摘要
根据黄酮类化合物能与 Al3 +形成稳定的荧光络合物 ,建立一种测定杜仲叶总黄酮的新方法。用 6 0 %乙醇加热回流提取杜仲叶有效成分 ,以芦丁为标样 ,选择激发波长 λex=4 36 nm,发射波长 λem=4 83nm,采用荧光分光光度法测定杜仲叶中总黄酮含量 ,并进行加标回收实验 ,验证方法的准确性。方法检出限为1.2 7× 10 -9mol·L-1,线性范围在 1.6 4× 10 -8— 3.6 3× 10 -5mol·L-1之间 ,线性回归方程 :y=17.92 3x+1.398,相关系数 r=0 .9989,平均回收率 99.8%— 10 4 .2 %。相对标准差 (RSD) 0 .84 % ,本法操作简便、快速、准确 ,具有良好的分析应用前景。
A new method for determining total flavanoid in leaves of eucommia ulmoide was e stablished based on the reaction of total flavanoid with alumimum ion to form fl uorescent materal.The sample was extracted with 60% ethanol by continuous therma l reflux,total flavanoid was determined by spectrofluorophotometer with rutin as standard reference,at the excitation wavelength of 436nm and the emission wavel ength of 483nm.The detection limit is 1.27×10 -9 mol·L -1 in the line ar range of 1.64×10 -8 —3.63×10 -5 mol·L -1 with the regression equation of y=17.923x+1.398(r=0.9989 and RSD=0.84%).The method is simple,ra pid,and accurate with a bright future in analytical applications.
