水牛乳腺上皮细胞的体外培养

Cultivation of Mammary Epithelial Cells from Buffalo

对水牛乳腺上皮细胞进行了原代培养，培养液由４０％的１９９、４０％的ＤＭＥＭ加２０％的犊牛血清组成。乳腺组织块接种在铺有鼠尾胶原的培养瓶中，在ＣＯ＿２培养箱中３７℃封闭培养一周后，可分生出多边形单层上皮细胞，培养过程中，成纤维细胞逐渐减少，二周后呈现以上皮细胞为主的不规则生长区域。在培养２５天的单层上皮细胞中，出现拉网（ｎｅｔｔｉｎｇ）结构。以上结果表明，利用乳腺组织块在体外培养可获得较纯的单层乳腺上皮细胞。

Primary culture of mammaryy epithelial cells from buffalo is performed with theculture fluid consisting of 40%of 199,40%of DMEM and 20%of FCS. Franments of mammarytissues are inoculated in a culture bottle containing rartail collagen and in the culture box at 37℃thesingle-layer epithelial cells proliferate from the fragments in a weekand two weeks later some irregulargrowth areas maiuly contxining single-layer epithelial cells are formed with the decrease of fibre cells.In the single-layer epithelial cells cultured for 25 days,netting structures are observed.The resultshows that relatively pure single-layer epithclial cells can be obtained by means of the cultivation ofmammary tissues.
