摘要
通过发芽块茎热处理(37℃)、茎尖(0.1~0.5mm,带1~2个原基)生长点剥离、分生组织培养(2~6个月)技术,配合血清学、电镜技术检测筛选,获得了6个马铃薯品种无PVX、PVY、PLRV等主要病毒的脱毒试管苗。本法可用于马铃薯脱毒快繁供种体系扩繁应用或研究。
Using heat-treatment of potato sprouting tubers and of meristem culture and detecting with serological and common (routine)Electron Microscope(CEM)and Immunoelctron Microscope(IEM),the tissue cultured seedlings of six potato varieties were obstained.The seedlings were free from main potato viruses such as PVX,PVY,PLRV etc and could be applied into virus-free potato seedling production in network.The tempreture of heattreatment was 37℃ and it lasted 4-15weeks,the length of stem-tips was 0.1-0.5cm(with 1 or 2 origin basses of leaf).The culture lasted 2-6 months.Methods of ELISA,CEM ,IEM were used in identification of the viruses.
