摘要
以云南芥菜型油菜部分品种及雄性不育系S187A、恢复系S187C为材料,进行花药培养诱导胚状体试验。结果表明,培养效率受材料基因型的影响较大;采用液体漂浮分步培养法较固体培养法诱导频率高,前者的胚状体诱导频率达42.1%,而采用固体培养方法仅为3.2%。在液体漂浮分步培养程序中加入适当浓度的多效唑,有利于提高胚状体的诱导频率,其中有的处理胚状体诱导频率达到214%。
Sterile S187A, restorer S 187C and some Yunnan local cultivars of Brassica juncea were used as materials for anther cultural experiments to induce pollen embryoids. Result showed that the inductive frequencies were somewhat influenced by different genotypes. stepped float anther culture (SFAC) had higher inductive frequency (42.1%) than that of solid culture (3.2%). By adding PPP333 in a favorable concentration into SFAC medium, it was helpful to increase the inductive frequencies of embryoids, perhaps it would be reached 214%.
