The effect of basic fibroblast growth factor on the neurons apoptosis after tractive spinal cord injury in rats

摘要 目的:探讨碱性成纤维细胞生长因子(bFGF)对牵张性脊髓损伤后神经元凋亡的影响。方法:大鼠脊髓T13-L2经牵张损伤,皮层体感诱发电位(CSEP)监测P1-N1波幅下降至术前波幅的70%后,于损伤平面以下经蛛网膜下隙置细导管,治疗组分别于术后即刻、0.5、1、2、3、4、8、12及24h经细导管注入bFGF溶液20滋l(含bFGF20滋g),对照组在相同时间注入等量生理盐水,然后于术后1d、4d、7d、14d及21d处死取材(n=4)。采用流式细胞仪及TUNEL法观察脊髓细胞凋亡情况,采用行为学评分及CSEP检查大鼠功能恢复情况。结果:术后4、7、14、21d,bFGF治疗组用流式细胞仪检测的凋亡率及TUNEL法染色示细胞凋亡均明显低于对照组(P<0.05、0.01)。神经行为学评分及皮层体感诱发电位也显示出大致相同的趋势。结论:碱性成纤维细胞生长因子能抑制牵张性脊髓损伤后神经元凋亡,并能促进脊髓功能恢复。 Objective:To explore the effect of basic fibroblast growth factor on neurons apoptosis after tractive spinal cord injury in rats.Method:With decreasing of the P1-N1 wave control 70% through CSEP monitor, the T13-L2 spinal cord of rats were injured by traction, and a thin plastic tube was situated in subarachnoid space below the injured level for perfusion. The bFGF-treated rats were received bFGF 20?滋g(20?滋l liquid)from the tube at once,30min,1,2,3,4,8,12 and 24 hours after injury respectively.The saline-treated rats were received the equal volume of normal saline at the same time, then rats were killed in 1?4?7?14 and 21 days postoperatively(n=4). The spinal cord cell apoptosis was observed with flow cytometer and TUNEL method. The functional recovery of rats was tested with combined behaviornal score(CBS) and cortical somatosensory evoked potential(CSEP).Result:Cell apoptosis ratio tested by flow cytometer and positive cells stained by TUNEL method were much lower in bFGF-treated group than in control group in 4?7?14?21d postoperationly(P<0.05,0.01). The trend of combined behavioral score(CBS) and cortical somatosensory evoked potential(CSEP) was also approximately the same.Conclusion:bFGF can inhibit neuron apoptosis and improve functional recovery of spinal cord after tractive spinal cord injury
