摘要
目的 探讨雷公藤多甙对小鼠髓样单核白血病细胞系WEHI -3B作用机理。 方法 应用四甲基偶氮唑蓝(MTT)法、流式细胞仪 (FCM )、电镜等方法检测雷公藤多甙对WEHI -3B细胞的影响。 结果 雷公藤多甙能显著抑制小鼠白血病细胞的增殖 ,降低细胞存活率 ,其半数细胞抑制剂量 (IC5 0值 )为 2 μg/ml;FCM检测雷公藤多甙实验组凋亡细胞百分率显著上升 ;雷公藤多甙实验组细胞核固缩 ,染色质边集。 结论 雷公藤多甙能显著抑制小鼠髓样单核白血病细胞系WEHI -3B细胞的生长 ,促进细胞凋亡。
Objective To study the mechanisms induced by Glucosidorum Tripterygll Totorum in WEHI-3B cells. Methods The Survival rate of cells were tested by MTT assay. Apoptotic cells were observed by fluorescence microscopy and apoptotic rate of cells were tested by flow cytometry. The change of cell phases were analyzed by flow cytometry. Submicroscopic structures were observed by transmission electron microscope. Results It suggested that Glucosidorum Tripterygll Totorum could notably reduce the survival rate of WEHI-3B cells, IC50 was 2 μg/ml The rate of apoptosis cells were increased in experimental group. Glucosidorum Tripterygll Totorum could induce the change of Submicroscopic structures. Conclusion Glucosidorum Tripterygll Totorum could notably inhibit the growth and proliferation of WEHI-3B cells and could induce the apoptosis of WEHI-3B cells.
