摘要
目的: 研究生骨注射液对体外培养的大鼠成骨细胞VEGFmRNA表达的影响, 探讨其促进骨折愈合的分子机制。方法: 体外培养大鼠成骨细胞, 分别予不同浓度和时间的生骨注射液干预实验, 然后用RT PCR技术检测各组细胞的VEGFmRNA表达, 并行定量分析。结果: 不同浓度的生骨注射液连续干预至第 5d, 以 1mg/ml浓度条件下, VEGFmRNA表达最强, 干预 1 ~5d, VEGFmRNA的表达随时间逐渐增强。结论: 适当浓度的生骨注射液对体外培养的大鼠成骨细胞VEGFmRNA表达有明显促进作用, 其作用强度与时间有关。
Objective: To study the effect of Shenggu injection on mRNA expression of vascular endothelial growth factor (VEGF) in rat osteoblasts in vitro and elucidate the mechanism of it in promoting the healing of the fracture.Method:The rat osteoblasts were isolated and cutured in vitro and treated with Shenggu injection according to the protocol. The expression of mRNA of VEGF in the cells in every group was examined by reverse-transcriptase ploymerase chain reaction (RT-PCR).Result:The expression of VEGF mRNA peaked with the stimulation concentration of 1mg/ml when osteoblasts exposed to Shenggu injection on the 5^(th) day at variant concentration. From the 1^(st) to 5^(th) day after treatment with the concentration of 1mg/ml of Shenggu injection, the expression of VEGF mRNA was gradually increased with increase of the cutured time.Conclusion:Stimulation of Shenggu injection with reasonable concentration can increase significantly the expression of VEGF mRNA in rat osteoblasts in vitro, the levels of expression of VEGFmRNA are correlated with the stimulation time.
