摘要
目的 :研究伊犁贝母总生物碱 (FPA)对豚鼠的平喘作用及其作用机理。方法 :以乙酰胆碱 组胺引喘法观察FPA对豚鼠的平喘作用 ;制备豚鼠气管螺旋条标本 ,通过观察其张力变化研究FPA对离体气管平滑肌基础张力的影响及其组胺和卡巴胆碱对气管收缩的抑制作用。结果 :FPA在 10 ,2 0 ,4 0mg/kg剂量下连续灌胃给药 5d ,对乙酰胆碱 组胺所致的豚鼠哮喘具有显著的延长引喘潜伏期的作用 ;对豚鼠离体气管平滑肌的基础张力无显著影响 ,对组胺引起的收缩也无显著的抑制作用 ,对卡巴胆碱引起的收缩 ,FPA在 10mg/L浓度时开始呈现非常显著的抑制作用 (P <0 .0 1) ,对 6 μmol/L卡巴胆碱的IC50 为 14 7mg/L。FPA可浓度依赖性地使卡巴胆碱量效曲线向右平移 ,最大收缩张力无显著变化。结论 :FPA对豚鼠具有平喘作用 。
AIM:To investigate the antiasthmatic effect and its mechanism of total alkaloids from Fritillaria pallidifora Schrenk(FPA)in guinea pigs.METHOD:The antiasthmatic effect was studied by observing the latent period of asthma induced by acetylcholine chloride and histamine phosphate in guinea pigs.By measuring the tension of guinea-pig isolated tracheal strips in vitro,we observed both influence of FPA on the basal tension and effects of FPA on the contraction induced by histamine or carbachol.RESULT:FPA 10,20,40 mg/kg were treated(ig)for 5 days.The results showed that the latent period of asthma in guinea pigs was significantly prolonged.The influence of FPA on both the basal tension and the contraction induced by histamine of guinea pig isolated tracheal smooth muscle were not significant. FPA could inhibite isolated tracheal smooth muscle contraction induced by carbachol 6 μmol/L,and its IC (50) was 14.7 mg/L.After incubated with FPA,the concentration-response curves of carbachol were shifted to the right in a concentration-dependent manner,and the maximal contractile response was not changed significantly.CONCLUSION:The results showed that the antiasthmatic effect of FPA was very significant,and its mechanism was related to the competitive antiagonism of muscarinic receptor on guinea pig tracheal smooth muscle.
