摘要
目的探讨细胞间隙连接蛋白(connexin,Cx)43在良、恶性前列腺组织中的表达和分布情况,以及与整合素连接激酶(Integrin-linkedKinase,ILK)表达的相互关系。方法采用免疫组织化学SP法检测Cx43、ILK在7例正常前列腺组织(NP)、16例BPH及52例前列腺癌(Pca)组织中的表达。结果Cx43既定位于细胞膜上,又在细胞质中表达。Cx43的阳性表达率在Pca组织中为32.7%(17/52)显著低于NP组织的85.7%(6/7)、BPH组织的87.5%(14/16)(P<0.01;P<0.005)。Cx43在Pca细胞中的阳性表达率与肿瘤细胞的分化程度呈正相关(P<0.05),C+D期显著低于A+B期(P<0.05)。Cx43阳性表达与ILK的表达呈负相关(P<0.001)。结论Cx43异常表达于BPH、Pca细胞的胞浆。Cx43表达的增加或下降在BPH的增殖过程中或Pca的分化、进展中起重要作用。Cx43和ILK蛋白在Pca组织中呈现相反的的表达方式,提示在Pca细胞增殖动力学调节中协同促进了肿瘤的恶性进展。
Objective To investigate the expression and distribution of connexin(Cx)43 on the biologicalbehavior in non-neoplasma and malignant human prostate, and its relationship with Integrin-linked Kinase (ILK).Methods Expression of Cx43 and ILK were examined immunohisochemically in 7 normal prostate(NP)、16 BPHand 52 prostate carcinoma (Pca). Results Expression of Cx43 protein was localized in cell membrane and cytoplasm.The positive rates of Cx43 in Pca and NP、BPH were 37.2% (17/52) and 85.7% (6/7), 87.5% (14/ 16) (P<0.01; P<0.005)respectively; positively correlated with the differentiation stages in Pca (P<0.05). The positive rates of Cx43 in A+Bstage and in C+D stage were 42.4% (14/33) and 15.8% (3/19) (P<0.05) respectively. Expression of ILK was negativelycorrelated with expression of Cx43 (Pearson’R=-0.712,P<0.001). Conclusions Cx43 protein was abnormallylocalized in BPH and Pca cytoplasm, there were a marked increase in BPH proliferated process or decreased in Pcadifferentiation and tumor progression. The contrasting expression of Cx43 and ILK may indicate a cooperation whichactivate the reglution of Pca proliferation kineties and malignant progression.
