摘要
目的研究深低温保存的猪二尖瓣经去细胞处理后生物学特征的变化.方法在含细胞保护剂二甲亚砜的保护液中深低温保存3个月的猪二尖瓣经高渗冲击、胰蛋白酶、核酸酶等交替处理后,行组织切片和扫描电镜观察细胞外基质的变化,对血液相容性变化进行研究,在小鼠皮下移植2~8周后测定钙含量,并比较其生物力学特征的变化.结果经去细胞处理后瓣膜细胞外基质完整,而细胞成分被去除,小鼠皮下移植后钙含量降低,而血液相容性和生物力学性质改变不明显.结论深低温保存的猪二尖瓣叶经去细胞处理后,减少了瓣叶细胞成分,对减少异种二尖瓣移植物失功衰败,提高远期效果具有重要意义.
Objective To investigate the biological and biomechanical characteristics of cryopreserved porcine mitral valve which underwent cell extraction. Methods Cryopreserved porcine mitral valves stored for 3 months in deep hypothermia(-196℃) were processed with decellular treatment.The changes of morphological characteristics were observed under light microscopy and electron microscopy.Calcium content of the valve leaflets was measured after 2~8 week subcutaneous implantation in rats.Biomechanical evaluation was carried out on the cryopreserved mitral valves which underwent with and without decellular treatment. Results The valves of decellularized group had compete extracellular matrix,lower calcium content in the leaflet,and no marked changes in bloodcompatibility and biomechanical property. Conclusion Cryopreserved mitral valve xenograft with cell extraction lost their cellular components without destroy of extracellular matrix,which is very useful to avoid the failure of mitral valve xenograft transplantation and valuable to prevent their degeneration and calcification.
