摘要
目的应用对红细胞补体受体1(ECR1)分子容量检测的实验方法,研究其对系统性红斑狼疮(SLE)的临床价值。方法戊二醛固定红细胞,将3×109的红细胞定量“液相包被”于V型板中,依次加入抗ECR1单抗碱性磷酸酶标记的第二抗体及可溶性底物显色,移显色液于比色孔,405nm比色,计算其吸光度A值。结果正常健康人ECR1分子数明显高于SLE患者(P<0.01)。结论本方法能准确测定ECR1分子的浓度及表达的差异性,其变化与SLE的发生发展及转归有明显的相关性,故对该疾病的诊断及疗效的评价有重要的临床价值。
Objective To investigate the clinical significance of quantitative expression of complement receptor type 1 on erythrocytes (ECR1) in patients with systemic lupus erythematosus (SLE). Methods In each sample, 3×109 glutaraldehyde-fixed red blood cells (RBC) were analysed in V well microtitre plates. In each well mouse IgG conjugated to alkaline phosphatase and the substrate was added following three washes with S/BSA. The supernatant was transferred to a clean U microtitre plate to faclitate reading by a plate reader at 405 nm (A). Results The quantitative expression of ECR1 in patients with SLE was significantly lower than that of healthy individuals (P<0.01). Conclusion The amount of ECR1 can be accurately examined by this technique. Quantitative expression of ECR1 may play an important role in the pathogeneses of SLE, and it is significantly associated with the development of SLE. Quantitative expression of ECR1 may have clinical significance in diagnosing and evaluating the prognosis of patients with SLE.
