摘要
目的 探讨神经生长因子 (nervegrowthfactor, NGF)及其神经生长因子受体 (nervegrowthfactorreceptor,NGFR)对先天性巨结肠(Hirschsprung'sdisease, HD)的调控作用。方法 采用RT PCR技术检测新鲜标本正常对照肠管、痉挛段、移行段和扩张段各 22例NGF和NGFRmRNA的表达,分析并统计其与临床病理特点之间的关系。结果 22例正常对照肠管 21例NGFmRNA表达阳性(95.5% ),20例NGFRmRNA表达阳性(90.9% );痉挛段 5例NGFmRNA表达阳性(22.7% ),6例NG FRmRNA表达阳性(27.3% );移行段 9例NGFmRNA表达阳性(49.9% ),7例NGFRmRNA表达阳性(31.8% );扩张段 18例NGFmRNA表达阳性 (81.8% ),19例NGFRmRNA表达阳性 (86.4% )。NGF和NGFRmRNA正常肠管与扩张段肠管中表达高度一致;痉挛段和移行段均为NGFmRNA和NGFRmRNA低表达。结论 NGF和NGFR与HD病理形态有相关性;HD的痉挛段和移行段肠管NGF及NGFR表达异常,提示NGF和NGFR基因可能与HD的发生有关。
Objective To explore the expression of nerve growth factor (NGF) and nerve growth factor receptor (NGFR) genes in Hirschsprung's disease (HD).Methods The mRNA of NGF and NGFR in normal, spastic, transitional and dilated segment were detected by RT-PCR in 22 fresh samples of HD. The relationship between expression of NGF and NGFR genes and pathological changes in colons were analyzed.Results In normal control, NGF mRNA was positive in 21 out of 22 ( 95.5%), NGFR mRNA was positive in 20 ( 90.9%). Meanwhile, NGF mRNA was positive in 18 ( 81.8) and NGFR in 19 ( 86.4%) in dilated segment. However, positive rate of NGF mRNA was decreased to 9 in transitional zone ( 49.9%) and to 5 in spastic segment ( 22.7%). Similar to the expression of NGF gene, NGFR mRNA was reduced to 7 ( 31.8%) in transitional zone and 6 ( 27.7%) in spastic segments.Conclusions NGF and NGFR genes may play some roles in the genesis of HD.
