丁酸对人结肠癌细胞尿激酶型纤溶酶原激活剂及受体表达的影响

The effect of butyrate on the expression of urokinase plasminogen activator and its receptor in human colon cancer cells

摘要  目的 探讨丁酸对人结肠癌细胞尿激酶型纤溶酶原激活剂(u-PA)及其受体(u-PAR)表达的影响。方法 将人传代结肠癌细胞株SW1116接种于不含或含不同浓度丁酸(2、3、4、7、10 mmol/L)的培养基中,培养6、24、48、72 h后,收集细胞用 ELISA法测定胞质中 u-PA和 u-PAR的表达情况。结果 随着培养基中丁酸浓度的增加和培养时间的延长,经丁酸处理的 SW1116 细胞表达的 u-PA和 u -PAR与对照组相比显著减少,当丁酸浓度≥4 mmol/L和培养时间≥48 h时,两者表达不再增加,反而呈逐步下降趋势。结论 丁酸可抑制SW1116细胞u-PA和u-PAR表达。 Objective To investigate the effect of butyrate on the expression of urokinase (plasminogen) activator (u-PA) and its receptor (u-PAR) in human colon cancer cells. Methods The (human) colon cancer cell line SW1116 was cultured in medium without or with different concentration of (butyrate)(2 mmol/L,3 mmol/L,4 mmol/L,7 mmol/L and 10 mmol/L). After being cultured for 6 h,24 h,48 h and 72 h the cells were collected and homogenized to measure the expression of u-PA or u-PAR by ELISA. Results The expression of u-PA and u-PAR in butyrate-treated SW1116 cell was significantly inhibited in a dose- and time-dependent manner. When the concentration of butyrate was more than 4 mmol/L and cell culture more then 48 h, the expression of both u-PA and u-PAR did not increase any more, and even tended to decrease gradually. Conclusion Butyrate can inhibit the expression of u-PA and u-PAR in colon cancer cells.
