摘要
本文研究了α-氨基酸与水杨醛和醋酸铜定量生成席夫碱-铜(Ⅱ)配合物的最佳条件,提出了用氯仿将过量水杨醛还原的铜与席夫碱-铜(Ⅱ)配合物离心分离,然后用火焰原子吸收法在324.8um处测定水相中的铜,从而间接求出α-氨基酸的含量.
In this paper, various conditions have been studied for the indirect determination of α-amino-acids by flame atomic absorption spectrometry. The α- amino acids react with a greater deal of salicylaldehyde to yield Schiff's base in alkaline solution. Schiff's base can react quantitatively with Cu(Ⅱ) to form a Schiff's base -Cu(Ⅱ)Chelate. The surplus copper(Ⅱ ) ion is reduced into copper or copper(Ⅰ) by the salicylaldehyde. Copper or copper (Ⅰ) are separated with the aid of chloroform in the centrifuge tube, Copper and copper(Ⅰ) go into organic phase, Schiff's base -Cu(Ⅱ ) chelate go into aqueous phase. Therefore, α- amino - acids can be determined indirectly by the aid of determination of copper in the aqueous phase by flame atomic absorption spectrometry.
