摘要
目的 探讨腺病毒介导的野生型p53在前列腺癌基因治疗中的可能性及其机制。方法培养前列腺癌细胞系PC3M,转染腺病毒分导的野生型p53,检测其增殖和凋亡水平变化,并检测其bax蛋白及mRNA水平。结果 腺病毒介导的野生型p53可高水平地转染人PC3M细胞,转染后的PC3M细胞增殖受到抑制、凋亡水平增高,细胞中bax蛋白及mRNA水平皆增高。结论 腺病毒介导的野生型p53可抑制PC3M细胞增殖并促进其细胞凋亡。其促进凋亡的机制可能是通过促进bax表达而实现的。
Objective To investigate the availability and mechanism of recombinant adenovirus vector expressing wild type p53(AdCMVp53) in gene therapy of prostate cancer. Methods Prostate cancer cell line PC3M was culti and revombinant adenovirus vector expressing wild type p53 was added to the. media. The levels of proliferation and apoptosis were detected and changes of bax were examined. Results In the transfect-ed cell with AdCMVp53 ,the level of proliferation increased and apoptosis decreased. There were also an increase in both protein and mRNA level of bax. Conclusions Recombinant adenovirus vector expressing wild type p53 could inhibit the proliferation of PC3M and promote its apoptosis. The mechanism of later may probably be its affection to bax.
