摘要 【目的】观察甲基莲心碱 (Neferine ,Nef)在体外对长春新碱 (Vincristine,VCR)抗人胃癌细胞(SGC790 1)作用的影响以探讨其化疗增敏作用。【方法】MTT法检测药物的细胞毒性 ,PI染色流式细胞术检测细胞凋亡。【结果】2 .5、5、10 μmol/LNef对SGC790 1细胞无明显细胞毒性 ,但可使VCR对SGC790 1的IC50 从 0 .0 5 9μg/ml下降至 0 .0 36、0 .0 30、0 .0 14 μg/ml。流式细胞术显示SGC790 1细胞的自然凋亡率为3.1% ,10 μmol/LNef与SGC790 1细胞作用 2 4h后 ,细胞凋亡率为 3.2 % ,说明Nef并不显著诱导SGC790 1细胞凋亡。 0 .5、2、4 μg/mlVCR与SGC790 1细胞作用 2 4h后 ,细胞凋亡率分别为 5 .80 %、6 .5 9%、16 .4 % ,VCR可一定程度的诱导SGC790 1细胞凋亡 ;当与 10 μmol/LNef合用时 ,其凋亡率明显增加 ,分别为18.5 %、2 3.7%、35 .4 %。【结论】Nef能增强VCR的体外抗肿瘤作用 ,可能是一种化疗增敏剂。 Objective To observe the effects of neferine (Nef) on the anticancer function of vincristine (VCR) against human gastric cancer cells (SGC7901) in vitro and explore its action of chemosensitization.MTT assay was used to detect the cytotoxicity of VCR and Nef in SGC7901 cells, while the apoptosis was checked by isoelectric point staining flow cytometry.Nef at concentration of 2.5 , 5 and 10 μmol/L displayed no obvious cytotoxicity to SGC7901 cells, but decreased the IC 50 of VCR to SGC7901cells from 0.059 μg/ml to 0.036,0.030 and 0.014 μg/ml, respectively. The natural apoptotic rate of SGC7901 cells was 3.1%, it still stayed in 3.2% after reaction of these cells with 10 μmol/L Nef for 24 h , implying that Nef did not significantly induce their apoptosis .After SGC7901 cells were induced by VCR at concentration of 0.5, 2, 4μg/ml for 24 h , their apoptotic rates were 5.80%, 6.56% and 16.4% ,respectively; indicating that VCR could induce apoptosis in certain degree. When VCR was added at these concentrations in combination with 10 μmol/L Nef, the apoptosis of SGC7901 cells increased to 18.5%, 23.7% and 35.4%, respectively. [Conclusion] Nef is able to potentiate the anticancer function of VCR in vitro and may be a chemosensitizer.
