摘要
采用高效液相色谱法测定虫草菌粉中核苷类成分,尿苷、鸟苷、腺苷及腺嘌呤能很好分离,其优化的色谱条件为:AlltimaC1 8(2 5 0mm×4 .6mm ,5 μm)色谱柱,乙腈-水(V/V =4 :96 )为流动相,流速为1 .0ml/min ,检测波长为2 6 0nm。该方法灵敏度高、定量准确、重现性好、回收率高(98.7%~1 0 1 .5 %) ,适合用于虫草菌粉中核苷类成分的分析。虫草菌粉中尿苷、鸟苷、腺嘌呤和腺苷的含量分别为1 0 .5 2mg/g ,8.5 6mg/g ,0 .70 8mg/g和1 .79mg/g ,其测定RSD范围为0 .5 1 %~1 .30 %(n =5 )。
A reversed-phase high performance liquid chromatography was developed for the determination of nucleotides (uridine,guanosine,adenosine and adenine)in cordyceps. The nucleotides were well separated and determined on an Alltima C18 column (250mm×4.6mm, 5μm)by using a mixture of acetonitrile-water (V/V=4:96) at 260nm with the flow rate of 1.0ml/min. The results showed that the developed method was sensitive, accurate, reproducible and high recovery(98.7%~101.5%) and suitable for analysis of nucleotides in cordyceps. The contents of uridine,guanosine,adenosine and adenine were ~10.52 mg/g,~8.56 mg/g,~0.708 mg/g and ~1.79 mg/g, respectively, with the average RSD ranged between 0.51% and 1.30% (n=5).
