摘要
目的 观察不同浓度胰岛素对HK- 2细胞TG- β1 表达的影响。方法 将不同浓度的胰岛素加入培养的HK- 2细胞中,用EL IS法测培养液上清液中TGF- β1 蛋白浓度,RT- PCR法测HK- 2细胞中TGF- β1 m RNA表达水平。结果 低浓度胰岛素不影响TGF-β1 表达;高浓度胰岛素在不增加TGF-β1 m RNA表达的情况下可增加HK-2细胞TGF-β1 蛋白分泌。结论 高浓度胰岛素可能在翻译后水平上调HK- 2细胞TGF-β1 表达,这种作用可能参与了糖尿病肾病肾间质纤维化过程。
Objective To examine the effect of insulin on thc expression of TGF -β_1 in HK-2 cell line.Methods Different concentra tions of insulin were added to cultured HK-2 cells.TGF-β_1 protein in supernatant was measured using s pe cific enzyme-linked immunosorbent assay(ELISA) while TGF-β_1mRNA expressi on was assessed through reverse transcriptase-polymerase chain reaction(RT-P CR) tec hnique.Results Low concentration of insulin did not aff ect the expression of TG F-β_1 while high concentration of insulin led to a time-dependent increase i n TGF-β_1 concentration in the cell culture supernatant.Conclusio n High concen tration of insulin can upregulate the expression of TGF-β_1 in HK-2 cells by post-transcriptional mechanism and this effect may be involved in the process of interstitial fibrosis in diabetic nephropathy.
