HPLC法测定不同产地川芎中川芎嗪的含量

HPLC Determination of Tetramethylpyrazine in Ligusticum chuanxiong Hort.

目的:测定不同产地川芎中川芎嗪的含量以控制其质量。方法:采用Xttera C_(18)(4.6 mm×250 mm,5 μm)色谱柱,以甲醇-10 mmol·L^(-1)醋酸铵(pH=9.05)-四氢呋喃(19:81:2)为流动相,流速为0.6 mL·min^(-1),柱温为室温,检测波长为281nm,以外标法测定了川芎药材中川芎嗪的含量。结果:川芎嗪在0.3—24μg·mL^(-1)(r=0.9995)的浓度范围内呈线性关系。川芎嗪的平均回收率(n=6)为98.3%。结论:该方法准确、简便、易行,可用于质量控制。

Objective:To assay tetramethylpyrazine in Ligusticum chuanxiong Hort.by HPLC for establishing a method of quality control.Method:An ODS column was used with a mobile phase consisted of mixture of methanol -10mmol L^(-1) ammonium acetate-tetrahydrofuran(19:81:2) at a flow rate of 0.6 mL·min^(-1) in this method, and UV detector wavelength was 281nm.Result:The linear range of tetramethylpyrazine was 0.3 -24μg·mL^(-1) (r=0.9995).The average recovery was 98.3% (n=6).Conclusion:An accurate, simple and rapid qualitative method to assay tetramethylpyrazine in Ligusticum chuanxiong Hort.was established.
