毛细管区带电泳对单唾液酸神经节苷脂(GM_1)的分离

摘要 目的:建立毛细管区带电泳法分离单唾液酸神经节苷脂(GM_1)。方法:采用熔融的硅胶毛细管柱(fused-silica capillary)90 cm×75μm(有效长度为83 cm),以含15 mmol·L^(-1)α-环糊精的100 mmol·L^(-1)硼砂缓冲液为运行缓冲液(pH 9.9),分离电压30 kV;检测波长:200 nm。结果:GM_1的2种组分分离良好。结论:本法可用于GM_1的2种组分的分离测定。 Objective: To establish a method using capillary zone electrophoresis for separating two forms of monosialogangliosides(GM_1). Method;The separation was performed on a fused-silica capillary 90 cm×75 μm(efficency length 83 cm). 100 mmol·L^(-1) borax buffer containing 15 mmol·L^(-1)α- cyclodextrin(pH 9. 9)was used as running buffer. The separation voltage was 30 kV. The UV detection wavelength was 200 nm. Results; The two forms of GM_1 were successfully separated. Conclusion:This method can be applied to the seperation and the assay of two forms of GM_1.
