摘要
目的 观察具有补气、活血、化浊作用的中药复方延肾口服液 (以下简称延肾 )对体外培养的人肾小球系膜细胞增殖及产生白细胞介素 - 8(Interleukin - 8,IL - 8)的影响 ,从细胞生物学水平探讨延肾防治慢性肾功能衰竭 (Chronicrenalfail ure ,CRF)的作用机制。方法 采用MTT比色法和双抗夹心ELISA法 ,观察延肾大鼠血清对体外培养的人肾小球系膜细胞增殖和IL - 8生成的影响。结果 脂多糖 (Lipopolysaccharide ,LPS)可明显刺激人肾小球系膜细胞增殖和产生IL - 8,延肾大鼠血清对LPS刺激后人肾小球系膜细胞增殖和产生IL - 8有抑制作用 ,其抑制程度与药物浓度有一定的量效关系 ,即随药物浓度的增加对系膜细胞增殖和产生IL - 8的抑制率也相应增加。结论 延肾对LPS刺激后肾小球系膜细胞增殖和产生IL - 8有明显的抑制作用 ,提示延肾抑制系膜细胞增殖和自分泌IL - 8是预防肾小球硬化发生发展的重要作用机制之一。
Objective We observed the effect of Yan Shen,a traditional Chinese drug having the actions of discharging turbid pathogens,invigorating qi and promoting blood circulation,on proliferation of human mesangial cells(huMsC) cultured in vitro and production of interleukin 8(IL-8),with the aim of studying the mechanism of Yan Shen on prevention and treatment of chronic renal failure(CRF).Methods Rats were gavaged with Yan Shen.The effects of rat serum on proliferation of huMsC and the production of IL-8 were observed by MTT colourmetry and ELISA.Results Proliferation of huMsC and production of IL-8 were significantly stimulated by lipopolysaccharide, whereas were significantly inhibited by medicated rate serum in dose-dependent manner.Conclusion Yan Shen significantly inhibits proliferation of huMsC and production of IL-8,which underlies its mechanism of preventing development of glomerulosclerosis.
