严重烫伤后大鼠小肠粘膜上皮细胞刷状缘二肽载体(PepT_1)的生物学功能改变 被引量:2

摘要 目的:探讨严重烫伤后大鼠小肠粘膜上皮细胞刷状缘二肽载体(PepT1)生物学功能的变化,以明确严重烫伤后肠上皮细胞蛋白质吸收的特点。方法:采用SD大鼠30%烫伤模型,于伤后0,1,3,5,7天断髓处死,取中段空肠制成外翻空肠囊,置于二肽溶液(10μCi/ml)中,比较烫伤大鼠与对照组大鼠肠上皮细胞对二肽的转运、摄取功能的变化。结果:严重烫伤大鼠小肠粘膜对二肽的转运和摄取功能明显下降(P<0.05),同时PepT1mRNA也下降。结论:严重烫伤可下调肠上皮细胞二肽转运载体mRNA的表达并使其对二肽的转运和摄取功能下降。 Objectives: To determine the changes of biological functions of dipeptide transporter (PepT 1) in normal and severe scalded rats. Methods: Male sprague-Dawley rats inflicted by 30% total body surface area (TBSA) III degree scalding were employed as the model. An everted sleeve of intestine 4cm long made by mid-jejunum was securely incubated in Kreb's solution with radioactive dipeptide (3H-Glycylsarcosine, 3H-Gly-Sar, 10uCi/ml) at 37℃ for 15min to measure the effects of uptake and transport of PepT 1 of small intestinal epithelial cell in normal and severe scaled rats. Results: The effects of transport and uptake of PepT1 in scalded rats were significantly decreased compared with controls (P < 0. 05 ). Conclusion : The effects of uptake and transport of dipeptide transporters in small intestinal epithelial cell with injury were decreased.
