摘要
目的 研究羟基磷灰石纳米粒子对骨肉瘤U2 OS细胞生长的影响。方法 通过体外细胞毒性实验、观察细胞形态及超微结构等方法 ,研究不同浓度羟基磷灰石纳米粒子对骨肉瘤U2 OS细胞生长的影响。结果 羟基磷灰石纳米粒子对骨肉瘤U2 OS细胞有明显的抑制作用 ,且随浓度 (31.2 5 μg/ml到 5 0 0 μg/ml)和作用时间 (1d到 3d)的增加而增加 ,细胞出现皱缩、核浓缩、核碎裂等凋亡特征。结论 羟基磷灰石纳米粒子抑制骨肉瘤U2 OS细胞的生长 ,在一定范围内随浓度和作用时间的增加而增加。
Objective To investigate the effect of hydroxyapatite nanoparticles (nmHA) on the growth of osteosarcoma U2-OS cell line.Methods The effect of nmHA at different concentrations on the growth of osteosarcoma U2-OS cell line was investigated through the observing of morphology,ultrastructure and cytotoxicity test.Results NmHA inhibited the growth of the cells,the inhibitive effect increased with the increasement of the concentrations (from 31.25 μg/ml to 500 μg/ml) and durations of time (from 1 day to 3 days).The trace of apoptosis with nuclear chromatin condensation and fragmentation as well as cell shrinkage were observed.Conclusion Hydroxyapatite nanoparticles inhibit the growth of human osteosarcoma cell line U2-OS,and the effect increases with the increasement of the concentrations and durations of time in certain extent.
