RASSF1基因在肝外胆管癌组织中转录表达及其临床意义的研究 被引量:14

摘要  目的 探讨RASSF1基因3种不同转录本在肝外胆管癌中的表达及其临床意义。方法用RT PCR的方法检测 48 例肝外胆管癌组织及 12 例癌旁正常组织中 RASSF1A、RASSF1B 和RASSF1C mRNA的表达情况。结果 RASSF1A 在肝外胆管癌组织中的转录表达缺失高达68 75%,其缺失与肝外胆管癌的淋巴转移(P< 0 05)及 TNM 分期(P< 0 01)相关。RASSF1B、RASSF1C的表达与肝外胆管癌的组织学类型、分化程度、淋巴转移不相关。结论 RASSF1 基因 3种转录本在胆管癌组织中的表达存在明显差异,转录本 RASSF1A与肝外胆管癌淋巴转移及 TNM分期相关,是一种肝外胆管癌的候选抑癌基因。 Objective To investigate the express of 3 different RASSF1 transcripts and its clinical significance in extrahepatic bile duct carcinoma. Methods The mRNA express of RASSF1A,RASSF1B and RASSF1C was detected by RT PCR in 48 extrahepatic bile duct carcinomas and 12 matched normal tissues. Results The rate of express loss of transcript A in extrahepatic bile duct carcinoma is 68 75%,and it was positively related with lymph node metasis(P<0 05) and TNM stages(P<0 01). RASSF1B and RASSF1A mRNA expression have no statistical relation with extrahepatic bile duct carcinoma. Conclusion There is a significant express difference among the 3 different RASSF transcripts in extrahepatic bile duct carcinoma.RASSF1A is closely associated with lymph metastasis and TNM stage of extrahepatic bile duct carcinoma,should be a new tumor suppressor gene.
