摘要
目的:建立反相高效液相色谱法测定血浆中阿普唑仑浓度的方法。方法:以乙腈 50mmol/L磷酸 盐缓冲液pH=6.0(35∶65V/V)为流动相,三唑仑作内标,在紫外波长220nm处检测。结果:血浆中阿普唑仑的 最低检出浓度为2μg/L,平均回收率为90.82%,线性范围10~200μg/L(r=0.9998)。日内和日间差异(RSD) 分别小于3%和4%。结论:反相高效液色谱法灵敏、快速、准确,可满足临床治疗监测和药代动力学的研究。
Objective: To develop reverse HPLC procedure for the determination of alprazolam in human plasma. Methods: The mobile phase consisted of 50 mmol/L potassium phosphate (pH6.0)-acetonitrile (65∶ 35,V/V) and UV detection wavelength was set at 220nm. Results: The lowest detection limitation of alprazolam was 2μg/L, and the mean recovery rate 90.82%. The linearity was 10 to 200μg/L (r=0.9998) and the relative standard deviations of within-day and between-day were less than 3% and 4% respectively. Conclusion: The method is rapid, sensitive and accurate enough for clinical therapeutic drug monitoring.
