摘要 目的:探讨神经母细胞瘤(NB)中酪氨酸激酶受体(TR Ks)和神经营养素(NTs)的基因表达及其意义。方法:采用逆转录聚合酶链反应(RT-PCR)法半定量检测27例N B中TrkA、TrkB、TrkC及BDN F m R NA的表达。结果:TrkA的高表达率及总表达率在低分期组均明显大于高分期组(P<0.05),而其高表达水平与患者2年存活率之间具有明显的相关性(P<0.01)。TrkB的高表达率及总表达率在高分期组均明显大于低分期组(P<0.05),而BD NF的三种表达率在两组间比较则均无显著性差异(P>0.05),但这两者的共同表达率在高分期组则明显大于低分期组(P<0.05)。TrkC的表达通常伴有TrkA的表达,而这两者的共同表达率在低分期组则明显大于高分期组(P<0.05),但它们的表达之间并不存在着关联性趋势。结论:RT-PCR法检测酪氨酸激酶受体和神经营养素的基因表达对NB肿瘤的分期和预后评估具有重要的临床意义。 Objective: To study the expression of tyrosine-kinase receptors(TRKs) and neurotrophins(NTs) in human neuroblastomas. Methods: Expression of TrkA, TrkB, TrkC and BDNF mRNA was semiquantitatively detected by reverse transcription-polymerase chain reaction in 27 cases of neuroblastomas. Results: The high and total rate of TrkA were expressed in significantly lower-stage group compared to higher-stage group(P<0.05) and the high level of TrkA expression was correlated positively with the two-year cumulative-survival rate of these patients(P<0.01). The high and total rate of TrkB were expressed in significantly higher-staging group compared to lower-staging group(P<0.05) and the three rates of BDNF expression in the two groups weren't statistically different (P>0.05), but the co-expression ratio of TrkB and BDNF showed a remarkable significance in higher-staging group more than lower-staging group(P<0.05). Expression of TrkC was usually accompanied by expression of TrkA and the co-expression ratio between them had a statistical significance in the lower-staging group compared to the higher-staging group (P<0.05), but there was only a non-significant trend between TrkC and TrkA expression. Conclusion: RT-PCR detection of TRKs and NTs has an important clinical significance of tumor stage and outcome for patients with neuroblastomas.
