The Expression and Significance of Syndecan-1 and E-Cadherin Proteins in Human Sporadic Colorectal Carcinoma

摘要 目的 探讨Syndecan -1和E -Cadherin两种蛋白在散发性大肠癌中的表达及意义。方法 应用免疫组化SP法 ,检测 5 2例散发性大肠癌和 18例正常大肠黏膜组织中Syndecan -1和E -Cadherin蛋白的表达。结果 Syndecan -1和E -Cadherin在散发性大肠癌和正常大肠黏膜组织的阳性表达率分别为 65 3 8%、10 0 %和 61 5 4%、10 0 % ,两者在散发性大肠癌和正常大肠黏膜组织的阳性表达率差异均有显著性 (P <0 0 1)。Syndecan -1和E -Cadherin的低表达与散发性大肠癌的浸润深度、淋巴结转移、Dukes分期显著相关 (P <0 0 5 ) ,其中Syndecan -1的低表达还与肿瘤的分化程度密切相关 (P <0 0 5 )。Syndecan -1和E -Cadherin在散发性大肠癌的表达呈正相关。结论 Syndecan -1和E -Cadherin的表达下调可能在散发性大肠癌的侵袭转移中起着重要作用 ,联合检测Syndecan -1和E -Cadherin对散发性大肠癌的预后判断有参考价值。 Objective To investigate the expression and significance of Syndecan-1 and E-Cadherin proteins in human sporadic colorectal carcinoma(SCC). Methods Immunohistochemical SP method was used to determine the expression of Syndecan-1 and E-Cadherin in 52 cases of SCC and 18 normal colorectal tissues. Results The positive expression rates of Syndecan-1 and E-Cadherin proteins were 65.38%,100% and 61.54%, 100% in SCC and normal colorectal tissues, respectively, the difference of which between SCC and normal colorectal tissues was statistically significant(P<0.01).Significant correlation existed among the decreased expression of Syndecan-1 and E-Cadherin, the depth of tumor invasion, lymph-node metastasis, and Dukes stages(P<0.05). Furthermore, the decreased expression of Syndecan-1 was closely associated with the grade of SCC differentiation(P<0.05). There was a positive correlation between the expression of Syndecan-1 and E-Cadherin in SCC. Conclusion The results suggested that the decreased expression of Syndecan-1 and E-Cadherin may play an important role in the invasion and metastasis of SCC, and combination detection of Syndecan-1 and E-Cadherin may serve as index to predict SCC patient prognosis.
