摘要
目的 :观察中药WPY对重症急性胰腺炎 (severeacutepancreatitis,SAP )肺组织内IL 1 βmRNA表达的影响。方法 :SD大鼠 48只随机分为 4组 ,正常对照组、SAP模型组、WPY高 ,低剂量治疗组。采用牛磺胆酸钠胰腺被膜下注射诱导大鼠SAP模型 ,分别于术后 3、6、1 2小时检测血清淀粉酶 ,半定量RT PCR检测肺组织IL 1 βmRNA的表达。 结果 :造模组血清淀粉酶显著升高 ,WPY治疗后血清淀粉酶显著下降 ;正常对照组、造模组和治疗组肺组织均可见IL 1 β的表达 ,造模组较正常对照组表达高 ,且随时间 3、6、1 2h变化肺组织内IL 1 β表达呈递增趋势 ,用WPY后 ,随着药物剂量增加IL 1 β表达呈递减趋势。 结论 :中药WPY可以降低大鼠急性胰腺炎早期 3 ,6,1 2h血清淀粉酶和肺内IL 1 βmRNA的表达 ,从而提示WPY可以减轻细胞因子介导的全身炎症反应。
Objective:To explore the expression of IL 1β mRNA in lung of rats with severe acute pancreatitis(SAP) and evaluate the effects of the Chinese medicine(WPY) .Method:SD rats were randomized into four groups: normal control, SAP model without treatment by WPY, SAP modle with treatment by WPY high and low dose . SAP model was induced by injection of 3% sodium taurocholic acid into the subcapsular of pancreas. Serum amylase amylase of 3h, 6h, and 12h with SAP was measured. Expressions of IL 1β mRNA in lung were detected by semi quantitative RT PCR. Results: Serum amylase levels were significantly increased after the induction of SPA, reaching the peak at 12h, WPY can statistical significance decrease serum amylase levels with WPY treatment compared without WPY treatment. Expressions of IL 1β in the lung during SAP increased with time and decreased by WPY treatment with dose dependence. Conclusions:The WPY can decrease the expressions of IL 1β mRNA in lungs and therefore can weak the response of the inflammations caused by cytokines.
