铜绿假单胞菌IMP型金属β-内酰胺酶的基因研究 被引量:1

摘要  目的 研究铜绿假单胞菌产金属β- 内酰胺酶(metallo -β- lactamases,简称金属酶)的编码基因及亚型。方法 临床分离的铜绿假单胞菌用全自动微生物分析系统鉴定,纸片扩散法进行抗生素敏感试验,采用纸片协同法检测金属酶表型,PCR法扩增金属酶基因,并对PCR产物进行序列测定,确定亚型。结果 24株耐亚胺培南铜绿假单胞菌共检出产金属酶菌株7 株。该7 株细菌均扩增出IMP型金属酶,其扩增片段含500个核苷酸,核苷酸及氨基酸序列经GenBank分析,为IMP -1型金属酶(注册号:AY386702),但有3个位点的同义突变。结论 7 株临床分离的铜绿假单胞菌产生的金属酶为IMP -1亚型。 Objective To identify the genotype of the metallo-β-lactamases produced by 7 strains of Pseudomonas aeruginosa. Methods Pseudomonas aeruginosa were identified by VITEK-AMS,antimicrobial sensitivity tests were performed by K-B method and metallo-β-lactamases- producing bacteria was screened by the double disk synergy test.The encoding genes of metallo-β-lactamases were amplified by PCR.The purified PCR products were sequenced by ABI automated sequencer.Results Seven strains of metallo-β-lactamases-producing Pseudomonas aeruginosa were detected in 24 strains of Imipenem-resistant bacteria. The encoding genes of metallo-β-lactamases were amplified as IMP by PCR.Their PCR products had 500 nucleotides with no difference between eath other and their corresponding amino acid sequences were determinted as IMP-1 metallo-β-lactamases(GenBank Accession No.AY386702),but nucleotides sequences had three mutations.Conclusion The metallo-β-lactamases produced by the 7 strains of Pseudomonas aeruginosa were determinted as IMP-1 metallo-β-lactamases.
