摘要  目的 研究Caspase 3与化疗药物VM26联合对U251细胞系的作用,探讨基因治疗与化疗联合应用治疗胶质瘤的可能性。方法 脂质体包裹Caspase 3转染U251细胞,应用VM26对转染组与未转染组U251细胞进行治疗。通过细胞生长曲线、MTT、TUNEL观察Caspase 3高表达与VM26联合对U251的影响。结果 Caspase 3在U251细胞中获得高表达,Caspase 3高表达未能诱导U251细胞凋亡。VM26作用于转染Caspase 3的U251细胞凋亡比对照组显著增加,细胞存活率下降。结论 转染Caspase 3对U251细胞生长无抑制作用,但Caspase 3高表达的U251细胞对VM26的化疗敏感性显著增加。 Objective To investigate the effect of combination Caspase 3 with VM 26 on glioma cell line U251. Methods Caspase 3 gene was transfected into U251 by FUGENE, then transfected and non-transfected U251 were treated with the chemotherapeutic agent VM 26 .The effect of over-expression of Caspase 3 combining with VM26 on U251 was evaluated by cell growth curve,MTT and TUNEL technique. Results Caspase 3 was over expressed in U251 .Over expression of caspase 3 alone couldn't induce apoptosis in U251.The apoptosis induced by VM 26 in U251 transfected with caspase 3 was more significant than that in the controls. Conclusions There was no significant effect on U251 by transfecting Caspase 3 alone. Over expression of Caspase 3 could increase the sensitization of U251 to the chemotherapeutic agents induced apoptosis .
