叶片衰老抑制基因P_(SAG12-ipt)转入籼稻不育系中A的研究 被引量:2

摘要 以质粒pSG516(ipt)为目的基因,pHQ9(hpt和gus)为选择 标记基因进行共转化,以PSD-1000-氦气基因枪介导,将ipt、hpt和gus基因转化到籼型不育系中A的核DNA中,得到了98个潮霉素抗性再生植株系。经过GUS基因组织化学染色检测、PCR检测以及Southernblot杂交分析,证实了叶片衰老抑制基因PSAG12-ipt已经整合到水稻基因组中。 pSG516(ipt) and pHQ9(hpt and gus)were co-transferred into Zhong A (cytoplasmic male sterile indica) genomic DNA by the particle bombardment of Biolistic PSD-1000/He Particle Delivery System (BIO-RAD) with the plasmid DNA mixed to Gold Particle. Ninety-eight independent lines were obtained by selecting for hygromycin resistance. Results of GUS, PCR and Southern blot demonstrated that the leaf-specific senescence-inhibitor gene (P_(SAG12-ipt)) has been integrated into rice genome.
