摘要
为了改善由薄片外植体所诱导的愈伤组织的胚性能力,研究了氮源对巴西香蕉(MusaAAACavendishsubgroupcv.Brazil)薄片愈伤组织诱导的影响。取香蕉组培苗的假茎和球茎切成1mm左右的薄片为外植体,能较快诱导出愈伤组织。结果表明,当改良的B5培养基中x([NH+3])为1∶26时,诱导出愈伤组织的形4] [NO-态最好,且愈伤组织诱导率高达93 2%;愈伤组织中形成拟分生细胞团的能力最好。添加高浓度的椰子汁也能改善愈伤组织形成的质量,培养基中φ(椰子汁)=3 2%时,含拟分生细胞团数目最多,这些愈伤组织具有较强的器官发生能力。
To improve the embryogenic capability of the calli induced from micro-cross section explants cut from corms of in vitro plantlets of banana (Musa AAA Cavendish subgroup cv. Brazil), the effects of nitrogen source and x[NH^+_4]/[NO^-_3] ratio on the callus induction were investigated. The maximal callus induction ratio, 93.2%, and the best quality of the callus could be reached when x[NH^+_4]/[NO^-_3]ratio was 1∶26 in the modified B5 culture medium. Additive coconut milk could also improve the quality of callus. Those calli which contained more number of meristemoid-like cell groups, showed more potentiality to be organogenesis.
