铝对海马神经元电压依赖性钙电流的作用及其机制

摘要 目的探讨金属铝对急性分离的大鼠海马锥体神经元的作用及其机制。方法利用全细胞膜片钳技术研究大鼠海马CA1 区锥体神经元,观察其高电压激活钙通道电流 (IHVA)的变化。结果铝在低浓度范围 ( <0.2mM)内对IHVA的幅度具有抑制作用。8 Br cAMP并不能抵消铝在低浓度范围内对IHVA的抑制作用。结论铝对钙通道具有抑制作用可能是其导致神经毒性作用的原因之一;铝对IHVA幅度的抑制可能不是通过影响钙通道的磷酸化过程而起作用的。 Objective To explore the effect of aluminium(Al) on hippocampal neurons. Methods Whole cell patch clamp technique was used to investigate the effect and mechanisms of aluminum on high voltage-dependent calcium current (I HVA) on acutely isolated hippocampal CA 1 pyramidal neurons in rats. Results The results were as following: ① In the low concentration (<0.2mM) Al reduced I HVA. ② 8-Br-cAMP could not cancel the reduction of I HVA when bath application of Al with low concentration. Conclusion The reduction of I HVA by Al may be one of the reasons for its neurotoxicity and this effect can' t involve the interactions with phosphorylation sites of VDCC.
