摘要
目的:根据黄酮类化合物能与Al3+形成稳定的荧光络合物,建立一种测定银杏叶总黄酮的新方法。方法:用60%乙醇加热回流提取银杏叶有效成分,以芦丁为标样,采用荧光分光光度法,选择激发波长λex=436nm,发射波长λem=483nm,测定银杏叶中总黄酮含量,并进行加标回收实验,验证方法的准确性。结果:本方法检出限为1.27×10-9mol·L-1,线性范围在1.64×10-9~3.63×10-5mol·L-1之间,线性回归方程:Y=1.9313X+0.2705,相关系数r=0.9989,平均回收率99.8%~104.2%,相对标准差(RSD)1.94%。结论:本法操作简便、快速、准确,具有良好的分析应用前景。
Objective:On the basis of reaction of total flavanoid with alumimum ion to form fluorescent material, the new method of determining total flavanoid in leaves of Ginkgo biloba L. was established.Methods:Using 60% ethanol by continuous themral reflux to extract the Ginkgo biloba L. leaves,taking rutin as standard sample, selecting the excitation wavelength 436 nm and the emission wavelength 483 nm with spectrofluorophotometer, determing content of total flavanoid, then inspect the accuracy by recovery test.Results:The detection limit was 1.27×10 -9mol·L -1,the range of linearity with concentration of rutin was between 1.64×10 -8~3.63×10 -5 mol·L -1,linearity relationship Y=17.923X+1.398, correlativity was 0.998 9, RSD was 0.84%.Conclusion:The method is simple, rapid, accurate and has bright future on analysis application.
