斑点免疫金银染色试验与斑点ELISA诊断血吸虫病的比较

COMPARISON OF DOT-IMMUNOGOLD-SILVER STAINING AND DOT-ELISA FOR DIAG NOSING SCHISTOSOMIASIS JAPONICA

以日本血吸虫成虫抗原，应用斑点免疫金银染色法（Ｄｏｔ－ＩＧＳＳ）和Ｄｏｔ－ＥＬＩＳＡ诊断日本血吸虫病，５５例粪检阳性病人血清抗体检出率均为１００．０％，５０例正常人血清阴性符合率均为９８．０％，与５０例肝吸虫病人的交叉反应分别为２．０％和４．０％，两法未见显著性差异（Ｘ１１＝０．３４４Ｐ＞０．０５）。但进一步研究显示金标法的混合阳性血清最终滴度较酶法高４～５倍；使用抗原节省；２０份阳性血清抗体检出阳性终点也较酶法高２～４个滴度。

Dot-immunogold-silver staining(Dot-IGSS)and Dot-ELISA were compared for diagnosig schistosomiasis japonica with adult-worm antigen.The results showed that the positive rates of the 2 methods in testing the sera from 55 pr oven cases were both 100%,the negative rates in 50 healthy persons were both 98.0%,and that the cross rates in 50 clonorchiasis patients were 2.0% and 4.0 % respectively.Between the 2 methods, the difference in sensitivity was insignificant(P>0.05).Further observations revealed,however, that the highest titer of the mixed positive sera was 4-5 times higher in Dot-IGSS than in Dot-ELISA, and hence the amount of antigen requircd would be less in the former than in the latter.It was also found that the titers in 20 schistosomiasis cases were 2-4 times higher in Dot-IGSS that in Dot-ELISA.
