摘要
目的 :研究IL -1受体拮抗剂 (IL -1ra)对IL- 1β介导的细胞生物学功能的影响 ,为基因工程药物IL- 1ra用于妇产科临床提供理论依据。方法 :取培养 3~ 5代的子宫内膜异位症的基质细胞分别加入不同浓度的IL -1β与IL- 1ra ,用ELISA双抗体夹心法测定细胞培养上清液中RANTE的含量。结果 :IL -1β作用后 ,EMs在位内膜基质细胞培养上清液中的RANTES含量明显增高 (P <0 .0 1)。当用一定浓度的IL 1ra封闭IL -1受体后加用IL -1β ,细胞培养上清液中RANTES含量呈降低趋势 ,结果有统计学意义 (P <0 .0 5 )。 结论 :IL- 1ra有拮抗IL- 1β诱导EMs在位内膜基质细胞分泌RANTES的作用。
Objective:To investigate the effects of interleukin-1 re ceptor antago nist(IL-1ra) on the production of RANTES induced by interleukin-1beta(IL-1β) in endometrial stromal cell of endometriosis.Methods:The endomet rial stromal cell of endometriosis cultured within 3~5 passage were exposed to various concentrat i ons of IL-1β with or without pre-treatment with IL-1ra.RANTES concentrat ion in the cell culture supernatant was measured with a sandwish ELISA (enzyme link ed immunosorbent assay).Results:RANTES concentration was high ly increased in the cells with IL-1β treatment alone(P<0.01).Whil e pretreating the cells with IL-1ra at a c ertain concentration,RANTES protein secretion in the culture medium was reduced si gnificantly(P<0.05).Conclusion:IL-1ra can inhibi t the IL-1β-induced R ANTES production in endometrial stromal cell of women with endometriosis.
