目的:探讨人子宫内膜腺上皮细胞中是否存在cAMP激活氯通道及前列腺素对该通道的调节作用。方法:全细胞膜片钳记录法。结果:在人子宫内膜腺上皮细胞中记录到一种forskolin(腺苷酸环化酶激活剂)激活的、非电压、非时间依赖性氯通道电流,其反转电位与Cl-平衡电位相近,并可被特异的氯通道阻断剂DPC抑制。PGE2也可激发氯通道电流,但其增加幅度明显低于forskolin激活电流。PGF2a不能激活腺上皮细胞的氯通道。结论:人子宫内膜腺上皮细胞存在cAMP激活氯通道,并可受PGE2刺激而开启。
Objective: To investigate the effects of forskolin, an adenylate cyclase activator, and prostaglandin E2(PGE2) on chloride current in human endometrial epithelial cells. Method: The chloride current was measured bywhole-cell patch clamp technique. Results: Forskolin activated a whole-cell current that exhibited a linear I-Vrelationship as well as time- and voltage- independent characteristics. The current was suppressed by the Cl-channel blocker (DPC) and reversed at -7.6±4.3 mV, close to the equilibrium potential for Cl-(-5.2 mV). Thechloride current can also be increased by PGE2, but not PGF2a. However the current magnitude induced by PGE2was significantly lower than that stimulated by forskolin. Conclusion: A cAMP-activated chloride channel wasconfirmed in human endometrial epithelial cells. It can be regulated by PGE2 and plays an important role in modulat-ing ion composition of human uterine fluid.
