α-生育酚对精索静脉曲张大鼠生殖细胞Bcl-2蛋白表达的影响及意义

The Effects of α-tocopherol on Bcl-2 Protein Expression of Spermatogenic Cells in Rats with Varicocele

摘要 目的 探讨α -生育酚对精索静脉曲张 (VC)大鼠睾丸生殖细胞Bcl-2蛋白表达的影响及意义。方法 采用成年雄性Wistar大鼠复制精索静脉曲张疾病模型 ,4周后开始每日肌注α -生育酚 0 5mg/ (10 0g体重 )共 4周 ,同时设手术对照组 (CG)及假手术对照组 (SOG) ;8周后应用免疫组化SP法检测睾丸组织中Bcl -2蛋白表达。结果 治疗组 (TG)动物左侧睾丸重量显著高于CG(P <0 0 5 ) ,低于SOG (P <0 0 5 ) ;TG睾丸生殖细胞Bcl -2蛋白表达率细胞凋亡率 [左侧 (75 82± 9 10 ) % ,右侧 (77 0 5±11 2 8) % ]显著低于SOG[左侧 (84 3 7± 9 64 ) %、右侧 (85 95± 10 13 ) % ] (P <0 0 5 ) ,而高于手术对照组 [左侧 (60 42± 12 5 7) % ,右侧 (63 3 6± 14 5 4) % ] (P <0 0 5 )。结论 精索静脉曲张大鼠生精细胞Bcl -2蛋白表达降低 ,α -生育酚能提高精索静脉曲张大鼠生殖细胞Bcl-2蛋白表达率 ,并可能通过Bcl-2蛋白通道的负向调节降低细胞凋亡 ,对精索静脉曲张导致的睾丸损害具有一定的保护作用。 Objective To explore the effects of α-tocopherol on Bcl-2 protein expression of spermatogenic cells in experimental varicocele rats. Methods Thirty adult male Wistar rats were randomly divided into 3 groups(n=10): treatment group (TG) was intramuscularly injected α- tocopherol (0.5mg/100g body weight, one time daily for 4 weeks) after establishing varicocele model by partial ligation of the left renal vein for 4 weeks, operation control group (OCG)and sham operation group (SOG). After 8 weeks Bcl-2 protein expression in the testes tissues was detected by immunohistochemical method. Results The weight of left testes in TG was obviously heavier than that in OCG(P<0.05), but significantly lighter than that in SOG. The positive rate of Bcl-2 expression of both testis spermatogenic cells in TG was significantly higher than that in OCG(P<0.05), and significantly lower than that in SOG(P<0.05). Conclusion The Bcl-2 expression of spermatogenic cells was down-regulated in varicocele rats, and α- tocopherol could increase the Bcl-2 expression in the spermatogenic cells damaged by varicocle, and had a certain protective effect for the testes injury resulted from experimental varicocele.
