摘要
目的:探讨全反式维生素A酸(ATRA)对体外培养小鼠囊胚转化生长因子β1 (TGF β1 )蛋白表达的作用,并了解其对着床前期胚胎发育的影响作用。方法:获取妊娠3.5 d小鼠囊胚,分别培养在含0,0.1,1,10μmol·L-1的ATRA的M199培养基中24 h,用免疫组织化学SP法,分别检测不同剂量组 ATRA对小鼠囊胚 TGF β1蛋白表达的状况。结果:ATRA可增强小鼠囊胚TGF β1 蛋白表达,表达强度呈现剂量依赖型。结论:ATRA对小鼠胚胎的发育具有细胞毒性作用,TGF β1 在小鼠胚胎发育的调节中发挥了重要的作用。
Objective: To study the effect of all-trans retinoic acid (ATRA) on the expression of TGF-β_1 protein in mouse blastocysts in vitro and its possible roles on the preimplantation mice embryo development. Methods: The blastocysts of 3.5 days old mice were exposed for 24 h to different doses of 0, 0.1, 1 or 10 μmol·L -1 all-trans retinoic acid and observed for the expression of TGF-β_1 proteins by immunohistochemical SP staining. Results: The expression of TGF-β_1 protein was up-regulated by ATRT in mouse blastocysts in a dose-dependent manner. Conclusion: This study examined the cytotoxic effect of ATRA on mouse embryos. TGF-β_1 may play critical regulating roles in mouse embryo development.
