咯利普兰对哮喘小鼠气道树突状细胞的作用

摘要 目的:研究选择性PDE4抑制剂(咯利普兰)对哮喘小鼠肺内树突状细胞(DC)数量的影响。方法:BalB/c小鼠32只随机分为4组,即对照组、哮喘组、氨茶碱组、咯利普兰组。采用免疫组织化学染色技术检测小鼠肺、气道DC;计算机图像分析技术对DC进行定量测定。结果:对照组小鼠的气管、支气管、肺泡和脏层胸膜构成了一个NLDC 145+DC网络,正常对照组肺内 NLDC 145+ DC 数量为(60±10)个·mm-2 上皮面积;哮喘组小鼠肺内NLDC 145+DC数量[(210±35.6)个·mm-2]较对照组显著增加(P<0. 01);哮喘小鼠经咯利普兰处理后,肺内DC数量明显低于未处理的哮喘组[(88±24.3)个·mm-2比(210±35.6)个·mm-2,P<0.01],并且选择性 PDE4抑制剂(咯利普兰)组与氨茶碱组比较,肺内DC数量亦显著下降(P<0.01)。结论:下调肺内 DC数量可能是选择性PDE4抑制剂与氨茶碱治疗哮喘的一个重要机制; 选择性 PDE4 抑制剂(咯利普兰)治疗哮喘既安全又有效,具有潜在的应用前景。 Objective: To investigate the effect of selective PDE4 inhibitor (rolipram) on the number of airway intraepithelial dendritic cells (DCs) in murine models. Methods: 32 BalB/c mice were randomly divided into four groups: control group,asthma group,aminophylline-treatment group and rolipram-treatment group. The animal models of asthma was established by OVA sensitizing-challenging,and 24 h after the last challenge,the pulmony and tracheal samples were extracted. Immunohistochemistry was used to detect DC,and computer image analysis were applied to analyze DC quantitatively. Results: After OVA challenge,the whole amount of dendritic cells in tracheal,bronchus and alveolar were revealed by horseradish peroxidase staining using the NLDC-145 monoclonal antibody. The number of NLDC-145 +DC in the bronchial mucosa of asthma mouse was more than that in healthy controls (P<0.01). After seven days treatment with rolipram,the number of NLDC-145 +DC was less than that in asthma group (P<0.01),and the number of NLDC-145 +DC of rolipram-treatment group was less than that in aminophylline-treatment group(P<0.01). Conclusion: It is suggests that dendritic cells are involved in the inflammation of airway of asthmatic murine and that rolipram may down-regulate the number of dendritic cells much more than aminophylline.
