摘要
目的:观察血管内皮生长因子(vascularendothelialgrowthfactor,VEGF)基因在兔股骨头滑膜细胞的表达,以期为股骨头缺血性坏死的基因干预提供实验依据。方法:实验于2002-12/2004-08在中国医学科学院昆明医学生物学研究所和解放军成都军区昆明总医院中心实验科完成。选择健康成年24周龄以上的日本大耳兔12只,对照组和基因组各6只。将重组VEGF基因的真核表达载体(pcDNA/VEGF)和空载体pcDNA各200μg分别注入基因组和对照组兔髋关节腔,采用免疫组化法检测pcDNA/VEGF和pcDNA在滑膜细胞中的表达。结果:VEGF基因转染7d后,可见pcDNA/VEGF在滑膜细胞高效表达,而对照组(空质粒pcDNA)则无。结论:重组VEGF基因可在兔股骨头滑膜细胞中表达,促进股骨头缺血坏死处新生血管形成和侧支循环建立,为基因干预股骨头缺血性坏死提供了理论基础。
AIM:To observe the vascular endothelial growth factor(VEGF) gene expresscon in synovial cells of femoral head in rabbits so as to provide an experimental basis for the gene intervention of avascular necrosis of femoral head.METHODS:The experiment was conducted in Kunming Institute of Medical Biology,Chinese Academy of Medical Sciences and the central laboratory in Kunming General Hospital of Chengdu Military Area Command of Chinese PLA from December 2002 to August 2004.Twelve Japanese flap eared rabbits over 24 weeks old were divided into control and gene group equally.The recombinant eukaryotic expression vector of human VEGF gene,pcDNA/VEGF(200 μ g) and zero vector,pcDNA(200 μ g) were injected into hip joint cavity of rabbits in gene and control groups respectively.The expression of pcDNA/VEGF and pcDNA in synovial cells was assessed with immunohistochemical method.RESULTS:At 7 days after VEGF gene transfection,the expression of pcDNA/VEGF was found high in the gene group,while the expression of pcDNA was not found in the control group.CONCLUSION:The recombinant VEGF gene expresses in synovial cells of rabbit femoral head,and the expression enhance the vascularization and the establishment of collateral circulation in avascular necrotic femoral head,which lays a theoretical foundation for the gene therapy for avascular necrosis of femoral head.
