人发角蛋白材料组织相容性及其对机体的影响

摘要 目的:评价人发角蛋白材料的组织相容性及对机体的影响。方法:实验于1998-05/2000-01在青岛大学人工材料实验室完成。选取健康Wistar大白鼠54只,随机分为3组,每组18只。将人工材料、正常人发、生理盐水制成悬液注入大鼠腹腔。15,30,45d时处死取静脉血离心,保留血清测定各项指标,取全血测大鼠血常规部分指标,及全血谷胱甘肽过氧化物酶(GSH-Px)和血清超氧化物歧化酶(SOD)活性的变化。结果:15,30,45d正常人发组白细胞分别为(4.8±1.6),(5.7±1.7),(3.1±0.5)×109L-1,生理盐水组分别为(2.1±0.7),(2.9±0.3),(2.6±0.5)×109L-1;正常人发组血小板分别为(271±87),(293±80),(147±43)×109L-1,生理盐水组分别为(56±16),(41±14),(50±11)×109L-1。两组比较,差异有显著性意义。而人发角蛋白人工肌腱组与生理盐水组比较差异无显著性意义。3组材料对心功能、肝功能及GSH-Px活性影响差异无显著性意义。对肾功能影响:生理盐水组无变化,其余二组开始升高,以后逐渐降至正常。至30d后,正常人发组与人发角蛋白人工肌腱组SOD活性均明显降低,与生理盐水组差异有显著性意义。45d后,除正常人发组SOD尚未恢复活性外,其余SOD均恢复活性。结论:人发角蛋白材料可使白细胞,血小板早期升高显著;使谷胱甘肽过氧化物酶也早期升? AIM:To evaluate the histocompatibility of the hair keratin artificial material and its effect on the organism.METHODS:The experiment was conducted in the laboratory for artificial materials of Qingdao University from May 1998 to January 2000.Forty five healthy Wistar rats were randomly divided into 3 groups,including 18 rats in each group.Hair keratin artificial material,healthy hair and normal saline were made into suspension and then injected into the abdominal cavity of rats respectively.At 15,30 and 45 days,the rats were sacrificed and the venous blood of them was centrifugated.The blood serum was remained to determine all the indexes of serum.The whole blood was drawn to measure the partial indices of blood routine.Meanwhile,the changes of glutathione peroxidase(GSH Px) of whole blood and activity of serum superoxide dismutase(SOD) were also determined.RESULTS:At 15,30 and 45 days,the white blood cells(WBCs) were (4.8 ± 1.6),(5.7± 1.7),(3.1± 0.5)× 109 L- 1 in the normal hair group and (2.1± 0.71.6),(2.9± 0.3),(2.6± 0.5) × 109 L- 1 in the normal saline group,and the blood platelets were (271± 87),(293± 80),(147± 43)× 109 L- 1 in the normal hair group and (56± 16),(41± 14),(50± 11)× 109 L- 1 in the normal saline group,so there was significant difference in the number of WBCs and blood platelets between the two groups.However,those were insignificantly different bqetween the hair keratin artificial material group and normal saline group.The different materials affected the heart and liver function and the activity of GSH Px insignificantly.The kidney function was found no any change in the normal saline group,but increased in initial phase and then fell down to normal in the other tow groups.After 30 days,the activity of SOD in the hair keratin artificial material group and normal hair group were both significantly decreased,as compared with the normal saline group,then at 45 days,it recovered to normal except normal hair group.CONCLUSION:It is indicated that hair keratin artificial material can increase the number of WBCs and blood platelets obviously,and it also can increase the activity of GSH Px at the early stage,but gradually recovers to normal after 45 days.The number of blood platelets and WBCs may be one of the indexes of early organic rejection.
