摘要
目的:了解大鼠深Ⅱ度烧伤后不同时间、不同厚度皮片移植对细胞增殖的抑制因子-抑癌基因p53基因表达影响。方法:实验于2002-07/2003-08在第四军医大学西京医院烧伤外科实验室及基础部生物化学实验室完成。每只大鼠背部两处1%深Ⅱ度烫伤,行自体刃厚、全厚皮片移植,移植时间分别为:伤后即刻、伤后1周、伤后2周,在皮片移植术后的1,2,3,4周时间点取材,每个时间点5个标本。提取标本组织总mRNA,行p53基因及内参照PCR扩增,电泳后比较各时间点总光密度值。结果:在伤后即刻、伤后1周及伤后2周皮肤移植p53基因表达量为0.82±0.12,0.72±0.09,0.65±0.07,刃厚、全厚皮片移植后p53基因表达量为0.63±0.06,0.85±0.11,在不同移植时间组及刃厚与全厚皮片组间比较差异均有非常显著性意义(P<0.01),p53基因在大鼠的创伤愈合中表现出了与尽早皮片移植及移植物厚度上的正相关性。结论:p53参与了大鼠烧伤后皮肤移植后的创伤愈合,尽快及尽可能厚的皮肤移植有助于p53表达的增高,可能与减少瘢痕形成有关。
AIM:To investigate the effects of different time and thickness of skin graft on the expression of inhibitor antioncogene of cell proliferation,p53 gene in rats after deep second degree burn.METHODS:The experiment was conducted in the Laboratory for Biochemistry of Department of Basic Medical Sciences and laboratory of Department of Burns,Fourth Military Medical University of Chinese PLA from July 2002 to August 2003.Two burn injuries of 1% total body surface area(TBSA) were made in back of rats with deep second degree burn,and the split and full thickness skin grafts were designed immediately,1 and 2 weeks after burn injury.The species were obtained at 1,2,3 and 4 weeks after skin graft with 5 specimens at each time point.The total mRNA of skin species were extracted and then performed with amplification of the p53 gene and inner control RT PCR.The total absorbance(A) was compared respectively at diffrernt time after the electrophoresis of RT PCR production.RESULTS:After burn,the expression of p53 gene was 0.82± 0.12,0.72 ± 0.09,0.65± 0.07 immediately,1 ,2 weeks in skin graft group respectively,and 0.63± 0.06,0.85± 0.11 in split and full thickness skin graft groups respectively,so there were significant differences among different graft time,and between split and full thickness skin graft groups(P< 0.01),and the expression of p53 gene during wound healing were positively correlated with earlier time for grafting and thicker skin graft.CONCLUSION:p53 gene participates in the wound healing of burnt rats after skin graft.The much earlier skin grafting and the thicker skin graft can contribute to the increase of p53 gene expression,which is possible concern of the decrease of scar formation.
