摘要 目的:旨在探讨血管内皮生长因子(vascularendothelialgrowthfactor,VEGF)在肺癌组织中的分布及其表达水平。方法:对象为南通大学附属医院2000-07/2001-05肺癌手术中新鲜肺癌组织标本。采用免疫组织化学法分析了VEGF在38例肺癌组织的胞内定位及其表达状态;并测定癌、癌旁及远癌组织中总RNA水平,以反转录聚合酶链式反应分析组织中VEGFmRNA的表达水平。结果:38例肺癌组织中VEGF阳性表达率为76%,其中在Ⅲ期、Ⅳ期肺癌达90%,伴有淋巴结转移者为84%,癌组犤(1.44±1.06)ng/g组织犦总RNA水平高于癌旁组犤(0.63±0.24)ng/g组织犦和远癌组犤(0.50±0.27)ng/g组织犦,并与总RNA浓度呈显著正相关(P<0.01);基因扩增显示VEGF121和VEGF165为肺癌组织中表达的两个主要基因片段。结论:肺癌组织中VEGF呈过度表达状态,VEGFmRNA分析可以作为肺癌血管生成及转移可靠指标。 AIM:To investigate the distribution and expression level of vascular endothelial growth factor(VEGF) in lung cancer tissues.METHODS:The subjects were 38 fresh specimens of lung cancer tissues from the Affiliated Hospital of Nantong University from July 2000 to May 2001.The VEGF cellular locations and expression in the 38 specimens of lung cancer tissues were analyzed with immunohistochemical staining.The level of total RNAs in the 38 specimens was measured in cancer,peri cancer and distal cancer tissues,and then the levels of VEGF mRNA expression were analyzed by the reverse transcription polymerase chain reaction(RT PCR) assay.RESULTS:The positive rate of VEGF was 76% in 38 cases of lung cancer specimens,specially 90% in those at stage Ⅲ and Ⅴ ,and those with lymph node metastasis(84% ).The expression level of total RNA[(1.44± 1.06) ng/g]was significantly higher than that of patients' paracancerous tissues[(0.63± 0.24) ng/g] and distant lung tissues[(0.50± 0.27) ng/g].The level of total RNA was closely correlated with total RNA concentration of lung carcinoma(P< 0.01).It was shown in the gene amplification,VEGF121 and VEGF165 gene fragments were two major fragments in lung cancer specimens.CONCLUSION:The expression of VEGF is super high in lung cancer tissues.The analysis for VEGF mRNA in patients with lung cancer can be used as a reliable indicator for angiogenesis and metastasis of lung cancer.
