摘要
目的:通过建立兔创伤性肢体深静脉血栓形成动物模型,探讨深静脉血栓形成与血细胞变化及炎症反应的关系,为肢体静脉血栓形成的早期预防和康复干预提供实验基础。方法:实验于2003-10/2004-10在昆明医学院外科总教研室完成。随机将新西兰大白兔分为骨折固定组、创伤固定组、无创伤固定组和空白对照组,每组各30只。用不同能量击打兔左下肢近端外侧,建立兔肢体深静脉血栓形成动物模型,造模后第5,7,9天抽取兔耳缘静脉血1mL,用全自动血球计数仪检测红细胞、白细胞、血小板、中性粒细胞、血红蛋白。结果:骨折固定组红细胞、血小板、血红蛋白均低于其余3组(P<0.01),白细胞高于其余3组(P<0.01);骨折固定组、创伤固定组中性粒细胞高于无创伤固定组、空白对照组(P<0.05);骨折固定组、创伤固定组股静脉血栓发生率高于无创伤固定组和空白对照组(P<0.01)。结论:兔创伤性肢体深静脉血栓形成动物模型中,应用全自动血球计数仪能准确、快速对血细胞进行检查;兔创伤性肢体深静脉血栓形成与血细胞变化有关;深静脉血栓形成后可观察到炎症反应。
AIM:To establish the animal model of the traumatic deep venous thrombosis(DVT) in limbs of rabbits,and then investigate the relationship of DVT with the variety of blood cell and the inflammatory reaction so as to provide an experimental basis for the early prevention and rehabilitative intervention of limb venous thrombosis.METHODS:The experiment was conducted in the General Department of Surgery,Kunming Medical College from October 2003 to October 2004.Totally 120 New Zealand rabbits were randomly divided into fracture fixation group,trauma fixation group,non trauma fixation group and blank control group with 30 each.The model of rabbit DVT was established by hitting the rabbit lateral left thigh with different energy of trauma.Blood 1 mL was drawn from rabbit auricular veins to measure the number of red blood cells(RBCs),white blood cells(WBCs),platelet,neutrophil cells and ferrohemoglobin at 5,7 and 9 days by using automatic globulimeter.RESULTS:The RBCs,platelet,haemoglobin in the fracture fixation group was less(P< 0.01),WBCs more than those in the other groups(P< 0.01).The neutrophil cells in the fracture and trauma fixation groups were higher than those in the other two groups(P< 0.05).The incidence of venous thrombosis was higher in fracture and trauma fixation groups than in the other two groups(P< 0.01).CONCLUSION:In the rabbit model of limb traumatic DVT,the number of blood cells can be counted quickly and accurately with automatic globulimeter.There is relationship between the variety of blood cells and rabbit traumatic DVT.Inflammatory reaction can be observed after DVT.
