银杏药质体对小鼠脑缺血再灌注损伤的保护作用 被引量:4

摘要 目的:研究银杏药质体对小鼠脑缺血再灌注损伤的保护作用,并初步探讨其作用机制。方法:实验于2002-10/12在湖北省中医院动物试验中心进行。将60只SPF级昆明种小鼠随机分为假手术组、模型组、尼莫地平组、杏灵颗粒组、药质体高剂量组、药质体低剂量组共6组,每组10只。每组小鼠均按要求术前给药,建立小鼠脑缺血再灌注模型后迅速断头取双侧大脑皮质,分别测定超氧化物歧化酶(SOD)、谷胱甘肽-过氧化物酶(GSH-Px)和过氧化氢酶(CAT)活性,丙二醛含量。结果:模型组小鼠脑缺血再灌期脑组织SOD,GSH-Px和CAT的活性明显低于假手术组,而丙二醛的含量犤(34.92±8.88)μmol/g犦明显高于假手术组犤(21.62±5.44)μmol/g犦。预先给予银杏药质体可明显减轻SOD,GSH-Px和CAT的抑制,减少丙二醛的生成。该作用优于等总黄酮剂量的杏灵颗粒。结论:银杏药质体可通过抑制脂质过氧化反应而保护酶活性,减少细胞损伤。 AIM:To study the protective effect of compound of gingko on cerebral ischemical reperfusion injury,and probe into the mechanism.METHODS:The experiment was performed in Animal Trial Center of Hubei Hospital of Traditional Chinese Medicine between October and December 2002.Sixty SPF Kunming rats were randomly divided into six groups:sham operation group,model group,nimodipine group,Xingling ginkgo granule group,high dosage of compound of gingko group,and low dosage of compound of gingko group and ten rats for each group.The rats in each group were received drug before operation.Following cerebral ischemical reperfusion operation got both cerebral cortexes were collected.The superoxide dismutase(SOD),glutathione peroxidase(GSH- Px), catalase(CAT), and malondialdehyde(MDA)were measured in turn. RESULTS:Activity of SOD,GSH- Px,and CAT in rats of model group were remarkably lower than those of sham operation group, but the MAD content[(34.92± 8.88) μ mol/g] was remarkably higher than that in sham operation group[(21.62± 5.44) μ mol/g]. Preoffered compound of gingko could reduce the inhibiting of SOD,GSH- Px and CAT obviously,and inhibit the production of MAD.The function was better than xingling granules of the same dose of total chromocor.CONCLUSION:The compound of gingko can protect the activity of enzyme by restraining lipid peroxidation,so as to reduce the cellular damage.#
