摘要
目的: 旨在研究SD大鼠牙周的类成牙骨质细胞、成骨细胞和牙周韧带成纤维细胞表达骨钙素和碱性磷酸酶的情况,探讨区分此三种细胞的方法。方法: 体外分别培养12周龄SD大鼠第一磨牙牙周的上述三种细胞,利用计算机图像分析仪和免疫酶联仪对细胞分别进行骨钙素免疫组化染色和碱性磷酸酶活力定量分析。结果: 骨钙素在类成牙骨质细胞和成骨细胞中呈强阳性表达,在牙周韧带成纤维细胞中几乎不表达。成骨细胞碱性磷酸酶活性最强,在成纤维细胞中适中,在类成牙骨质细胞中几乎无活性。结论: 体外培养的SD大鼠三种牙周细胞表达骨钙素和碱性磷酸酶有各自特点,可以依此对三种细胞加以区分。
Objective:To compare the capacity of three types of periodontal cells of SD rat expressing osteocalcin and ALP and investigate the feasibility of distinguishing them in vitro. Methods: Osteoblasts,PDL fibroblasts,and cementoblasts-like of 12-week-old SD rat were respectively obtained from alveolar bone and the root surface of rat first molars and cultured.Quantitative immunohistochemistry study by computer and spectrophotometer were used to assess the osteocalcin(OCN) and ALP activity of three types of cells. Results: OCN was localized in osteoblasts and cementoblasts-like cultured in vitro.The highest level of ALP activity was found in osteoblasts,a moderate level in PDL fibroblasts.The cementoblasts-like lacked ALP activity. Conclusions: OCN can be immunolocalized on cultured cells to distinguish mineral-forming cells from fibroblasts. ALP can distinguish cementoblast-like from osteoblasts.This can serve for a simple mathod to distinguish the three types of periodontal cells cultured in vitro.
