摘要
目的研究转录因子NF-κB在肾移植急性排斥反应中的作用。方法ACI大鼠为供体,LEW大鼠为受体建立急性排斥动物模型,将移植肾分离制成核蛋白提取液,采用gelshift方法观察移植肾组织NF-κB与炎性基因启动子中特殊基因序列的结合能力,并以LEW大鼠同为供受体的移植肾作为对照。结果在急性排斥反应移植肾中,NF-κB的结合能力明显高于无排斥反应的同系移植肾。结论NF-κB在同种异体移植物中的强表达可能是急性排斥中的关键因素,提示可通过抑制NF-κB活性来降低移植物的排斥反应。
Objective To Investigate the function of NF-κB as a sequence-specific DNA binding protein in acutely rejected renal grafts. Methods Use gel shift assay to observe the NF-κB binding activity in renal grafts. Results DNA binding activity of NF-κB was enhanced in allografts significantly, whereas it almost could not be detected in isografts. Conclusion NF-κB play an important role in acute graft rejection. We suggest the interruption of NF-κB activity approach to prevent allorejection.
