摘要
目的 探讨环氧合酶 2(Cyclooxygenase 2,COX 2)在子宫内膜癌组织中表达及与肿瘤血管形成的关系。方法 采用免疫组化S P方法检测34例子宫内膜癌组织COX 2和血管内皮生长因子(Vascu lar endothelial growth factor, VEGF) 的表达和微血管密度(Microvessel density, MVD),观察COX 2表达与肿瘤血管形成之间的相关性。结果 COX 2在子宫内膜癌组织中的阳性表达率为64.7%,而对照组正常子宫内膜均未见表达,内膜癌组的中分化细胞 COX 2 蛋白的表达高于低分化细胞,差异有显著性(P<0.05);COX 2表达阳性组和阴性组MVD分别为(41.53±19.10和28.79±8.20),两组比较具有显著性差异(P<0.05)。COX 2的表达评分与VEGF及MVD高度均呈正相关(P<0.01;P<0.0001)。结论 COX 2可能主要参与子宫内膜癌发生的早期;子宫内膜癌组织中 COX 2 的高表达可能在 VEGF诱导肿瘤血管形成的过程中起重要作用。
Objective To investigate the expression of cyclooxygenase 2 (COX 2) in endometrial carcinoma and its relationship to angiogenesis.Methods The expression of COX 2 and vascular endothelial growth factor (VEGF) in 34 cases of endometrial carcinoma was evaluated by immunohistochemistry SP staining. Microvessel density (MVD) of endometrial carcinoma was also determined with anti CD34 as the label.Results The expression late of COX 2 in 34 cases was 64.7 % but not in control endometrium. The level of COX 2 expression was higher in grade Ⅱ tumor than in grade Ⅲ tumor ( P < 0.05 ). MVD was ( 41.53 ± 19.10 ) and ( 28.79 ± 8.20 ) in endometrium carcinoma positive and negative for COX 2 ( P < 0.05 ). The expression of COX 2 was positively correlated with expression of VEGF and MVD ( P < 0.01 ; P < 0.0001 , respectively ).Conclusion COX 2 may involve early in endometrium neoplastic progression. The expression of COX 2 in endometrial carcinoma may induce the expression of VEGF, increase angiogenesis and enhance tumor growth and invasion.
