摘要
目的 观察右侧迷路毁损后前庭内侧核 (medialvestibularnucleus ,MVN)早期快反应基因c -fos的表达特征。方法 选择健康白色赤目纯种豚鼠 2 7只 ,将豚鼠右侧迷路毁损后 ,在不同存活时间行前庭内侧核c -fos蛋白的二步法免疫组化染色 ,光学显微镜下观察染色结果。结果 对照组未见Fos样免疫反应。一侧迷路毁损 (u nilaterallabyrinthectomy ,UL)后可诱发双侧MVNFos样免疫反应 ,且术侧大于健侧。结论 ①UL毁损可诱导MVNFos样免疫反应增强 ,是前庭神经系统对外界刺激应答的一种方式 ,早期Fos样免疫反应 (1~ 6h)可能反应了复杂的神经通路的活性 ,延迟Fos样免疫反应 (2 4~ 4 8h)可能构成了前庭损伤后恢复过程中可塑性改变的基础 ;②前庭功能具有巨大的代偿潜力 ;③fos靶基因的本质及其他早期癌基因 (immediateearlygene,IEG)的作用仍待进一步研究。
Objective To investigate the c-fos ex pr ession in guinea pigs' medial vestibular neuleus (MVN) following unilateral laby rinthectomy (UL). Methods Twenty-seven SCO guinea pigs were randomly divide d into nine groups, three in each group. After resection of right labyrinth,th e immunohistological staining of c-fos was performed. Fos-like immunoreactivity (FLI) was analyzed at fifteen minutes, and 1?2?6?8?12 and 24 h, and 2 and 7 days after UL. Results Results showed no Fos expression in the control g roup. By contrast, Fos was consistently induced in the UL guinea pigs. Asymmetri cal labeling was found in the bilateral medial vestibular nucleus, with more Fos -like neurons in the ipsilateral MVN of the lesioned side than that in the contr alateral MVN(P<0.05). Conclusion ①UL can induce reactive strong Fos immunostai ning in the MVN, which is one of the important way in which vestibular neuron sy stem reacts to outside stimulation. By comparing these data to the behavioral re covery process, we conclude that the early Fos-LIR likely reflects the activatio n of neural pathways in response to UL whereas the delayed Fos-LIR might underli ne long-term plastic changes involved in the recovery process. ②The vestibular function has the great ability of compensation; ③The essence of c-fos and other IEGs' function need further research.
